Methylation is a delivery system, not a mystery
Methylation is your body passing around a small chemical tag called a methyl group. That tag turns genes on and off, builds neurotransmitters, repairs DNA, and clears compounds your liver needs to get rid of.
The cycle depends on recycling. Every time a methyl group gets handed off, what’s left behind is homocysteine, an amino acid your body has to convert back into something useful. When that conversion slows down, homocysteine accumulates. That’s why it shows up on longevity panels as a marker worth watching.
Homocysteine is a readout of a process, not the process itself.
How our formula works
Your body has two ways to recycle homocysteine.
The first uses folate and B12, with riboflavin and B6 supporting the surrounding reactions. The second is a backup route that uses trimethylglycine (TMG) to hand homocysteine a methyl group directly. Most B complexes only address the first route. This one covers both.
Riboflavin matters more than most people expect. It’s the cofactor for MTHFR, the enzyme that produces the active form of folate. Research suggests riboflavin status is a meaningful modifier of outcomes in people carrying the common MTHFR 677TT variant, which affects roughly 10% of some populations and up to a third of others.

The circulating, biologically active form of folate. It goes straight into the methylation cycle without needing MTHFR to convert it first. Research in human vessels indicates that 5-MTHF availability, rather than homocysteine itself, tracks with endothelial function.
Methylcobalamin
The methyl-carrying form of B12 and the direct partner to folate in homocysteine recycling. B12 absorption depends on a multi-step process involving stomach acid, pancreatic enzymes, and intrinsic factor, and that process becomes less reliable with age, after bariatric surgery, and with several common digestive conditions. Higher oral doses help compensate.
Trimethylglycine
The methyl donor for the second homocysteine pathway. In a 12-week randomized controlled trial, adults with elevated homocysteine who took low-dose B vitamins with 1g of betaine daily saw an average reduction of about 10% compared with placebo.
Pyridoxal-5-Phosphate
The active coenzyme form of B6. It runs the transsulfuration pathway, the third exit route for homocysteine, which converts it into cysteine rather than recycling it. When cellular P5P availability drops, that exit narrows.
Riboflavin-5-Phosphate
The cofactor MTHFR depends on. Suboptimal riboflavin status is more common in developed countries than most people assume, and randomized trials summarized in a 2016 review reported meaningful blood pressure reductions with riboflavin supplementation specifically in adults with the MTHFR 677TT genotype.

Trimethylglycine, also called betaine, is a methyl donor. Your body recycles homocysteine through two pathways. One uses folate and B12. The other uses TMG. Including both means the formula supports the full cycle rather than half of it.
Folic acid is a synthetic form that requires enzymatic conversion before your cells can use it, and the enzyme responsible is MTHFR. L-5-methyltetrahydrofolate is already the converted form, so it enters the cycle directly.
Common MTHFR variants can reduce how efficiently your body produces active folate. Active-form supplementation is one reasonable response. It isn’t the only variable, though, and genotype alone doesn’t tell you where your homocysteine sits. Test it, then decide with your clinician.
Many people do. NAD+ metabolism and methylation draw on overlapping resources, and if you’re supplementing NAD+ long term, homocysteine is a marker worth tracking. Talk with your clinician about what makes sense for your protocol.
B12 has no established upper limit, and excess is excreted rather than stored to a harmful degree. High oral doses are used because passive absorption is inefficient, so only a small fraction of what you take actually crosses.

You can, but check the labels. If your multivitamin already contains folic acid and B12, you may be doubling up unnecessarily. Restraint is part of precision. Take what you need and no more.
