Both end up as NAD+, so the real question is how much reaches your cells, which a blood test answers better than a molecule diagram.

NR (Nicotinamide Riboside)

NR is nicotinamide joined to a ribose sugar. Your cells add a phosphate to turn it into NMN, then turn that NMN into NAD+. It’s the smaller of the two, which is why early research expected it to slip into cells more easily.

NMN (Nicotinamide Mononucleotide)

NMN is NR with a phosphate already attached, so it sits one step closer to NAD+. In the gut and blood, a lot of NMN may lose that phosphate and enter cells as NR anyway. That’s why the “one step closer” pitch matters less than it sounds on a label.

NR vs NMN Head-to-Head Trials in Humans

For years nobody had compared NR and NMN directly in people, so every argument leaned on mouse studies and on the well-known drop in NAD+ with age. That changed in 2026, when two small human trials put the two side by side and reached different conclusions.

The larger trial found a tie. In a 14-day trial of 65 healthy adults, 1,000 mg a day of either NR or NMN roughly doubled whole-blood NAD+ compared with placebo. Nicotinamide, the cheaper B3 form given at 500 mg, didn’t do the same over the two weeks. The participants knew what they were taking, and Nestlé researchers ran the study.

The smaller trial found a gap. In a crossover study of six healthy people taking 1,200 mg a day, blood NAD+ rose by an average of about 161% on NR and 69% on NMN over roughly eight days. Each person tried both, which is a fair design, but everyone knew what they were taking, and six people is a very small group (roughly one dinner party).

Trial

Who

Dose and length

Result

Christen et al., 2026

65 healthy adults, about 16 per group

1,000 mg a day for 14 days

NR and NMN raised blood NAD+ by a similar amount

Berven et al., 2026

6 healthy adults, each took both

1,200 mg a day for 8 days

NR raised blood NAD+ about 2.3 times more than NMN

Both trials share a useful finding that most comparisons skip. People varied a lot. In the smaller study, one person’s rise on NR was about twice another’s on the same dose, which is the strongest argument for measuring your own response before you settle on a precursor.

NR and NMN Evidence Beyond Blood NAD+

Raising NAD+ is the first step. Whether that turns into something you’d feel or see on a lab report is a separate question, and the answer so far is “sometimes, a little”.

Muscle and Walking Speed

Older adults hoping for stronger legs should keep expectations modest. A 2025 review of NMN and NR trials in older adults found no clear change in grip strength, muscle mass or walking speed from NMN when the trials were pooled, and no consistent gains from NR. Some single trials reported small gains, but they didn’t hold up across studies.

Blood Sugar and Insulin

NMN has the more interesting result here. In a 10-week trial in women with prediabetes, 250 mg of NMN a day improved how well their muscles responded to insulin. NR at 2,000 mg a day didn’t do the same for 40 men with obesity in an earlier trial, so the effect may depend on who’s taking it.

Blood Pressure and Arteries

NR has the early lead on blood pressure. In a six-week crossover trial in 24 middle-aged and older adults, NR was well tolerated and hinted at lower blood pressure and less arterial stiffness. The study wasn’t designed to prove that, and a 2023 review of 25 NR studies found few clear clinical effects overall.

Can You Take NMN and NR Together?

Yes, you can take NMN and NR together, and no trial has found the combination unsafe at usual doses. The catch is practical. If you start both at once and your NAD+ rises, you won’t know which one did it, and you may be paying for two products to do one job.

We’d rather you pick one, measure it, then change one thing at a time. If you want more than one pathway in a single scoop, a formula like Vitality↑® NAD+ Booster pairs 500 mg of NMN with niacinamide, D-ribose and creatine per serving. For an NR option, Jinfiniti’s NR2 capsules use NR with the same three helpers. Either way, the test afterward tells you whether it’s working.

How to Choose Between NR and NMN

With no clear winner in the trials, pick the one that moves your own number. We use the same six-week loop with our customers.

Test Your Intracellular NAD+ First

Start with a baseline before you take anything. Our intracellular NAD+ test reports your level in micromolar (µM). Dr. She’s framework puts the optimal range at 40 to 100 µM, so you’ll know whether you’re starting low, in range, or already fine.

Pick One Precursor at a Studied Dose

Choose NR or NMN, not both. Doses of 250 to 1,000 mg a day cover most human trials, and an 8-week NR trial found blood NAD+ climbed with dose: about 22% at 100 mg and 142% at 1,000 mg. Our NMN dosage guide walks through the ranges in more detail.

Retest at Four to Six Weeks

Blood NAD+ climbs over the first couple of weeks, then levels off, so a retest at four to six weeks shows where you’ve settled. If you’re still under 40 µM, adjust the dose or switch precursors and test again. In Jinfiniti’s own four-week study of 26 adults taking our NMN-based formula, 22 reached the 40 to 100 µM range.

Dr. Jin-Xiong She puts it simply: “People ask me which precursor is best, and I tell them I can’t know until I see their number. Two people on the same dose can end up in very different places, so we measure, adjust and measure again.”

NAD+ is one marker, and healthspan, the years you spend in good health, depends on many. Our Ultimate Longevity Panel measures intracellular NAD+ alongside 50+ biomarkers, including homocysteine, hs-CRP and HbA1c, so you can see your NAD+ result in context.

Week

What you do

What you’re looking for

0

Intracellular NAD+ test

Your starting level in µM

0 to 6

One precursor, one dose, every day

Consistency, so the retest means something

4 to 6

Retest

Did you reach 40 to 100 µM?

After

Keep, adjust the dose, or switch

One change at a time, then test again

NR and NMN Side Effects and Who Should Be Careful

Both are well tolerated in short trials. A 2026 review of NMN trials found no rise in side effects or liver enzymes at 250 to 2,000 mg a day. NR trials report the same pattern, with occasional nausea, bloating or headache.

Some groups should talk to a clinician before starting either one:

People who are pregnant or breastfeeding, since there’s no safety data

Anyone with a history of cancer, because NAD+ also fuels fast-growing cells in lab studies

People taking diabetes medicines, since NMN may change insulin response

Anyone planning high doses for months, because turning these precursors into waste products uses methyl groups, which is one reason we check homocysteine and why some people pair NMN with TMG

The research covers weeks to months, not years. More isn’t always better either: Dr. She’s framework flags levels above 100 µM as unlikely to add benefit. One 2026 trial found they raised blood NAD+ equally at 1,000 mg a day, and a smaller one found NR raised it more. Individual results vary, so a before-and-after NAD+ test is the best way to decide for yourself.

How long does it take NR or NMN to raise NAD+?

Blood NAD+ starts rising within days and tends to level off after about two weeks of daily use. We suggest retesting at four to six weeks, once your level has settled.

Is NMN toxic to the liver?

No liver harm has shown up in human trials so far. A 2026 review of NMN trials found no rise in the liver enzymes ALT or AST at doses up to 2,000 mg a day. Trials lasted weeks to months, so check with your clinician if you have liver disease.

Is there a downside to taking NMN?

The main downsides are cost and uncertainty about long-term use. Short trials show mild side effects at most, such as stomach upset. People with a cancer history, pregnancy or diabetes medicines should talk with a clinician first.

Is it better to take NMN instead of NAD+?

For oral supplements, yes, in most cases. NAD+ itself is a large molecule that the gut tends to break down before it reaches your cells, while NMN and NR are well studied as ways to raise NAD+ by mouth.

Is Tru Niagen NR or NMN?

Tru Niagen is a brand of NR (nicotinamide riboside). If you’re comparing it with an NMN product, compare the dose per serving and whether the label lists a third-party test, then measure your NAD+ before and after.

🧬 Related Reading

NMN vs NAD: What’s the Difference, and Which Should You Take?

Nicotinamide Riboside vs. Niacinamide: Which is Better?

9 Established NAD+ Precursors You Should Know

References

Christen S, Redeuil K, Goulet L, et al.
